ABSTRACT

Objective@#Coronavirus disease-2019 (COVID-19) pandemic-related psychological symptoms can lead to smartphone addiction (SPA) risk and other behavioral disorders, thus impacting individuals’ mental health and well-being. The present study aims to propose a mediation model to investigate the association between emotional intelligence (EI) and SPA, and the mediating role of future anxiety (FA) during the post-COVID-19 era. @*Methods@#An online questionnaire including the Emotional Intelligence Scale, the Dark Future Scale, and the Smartphone Addiction Scale among university students from China, was conducted between September 14 and November 22, 2022. Finally, 1,154 valid questionnaires were collected. The reliability and confirmatory factor analysis results showed that all three scales had good reliability and validity. @*Results@#Structural Equation Model demonstrated that EI significantly and negatively influenced SPA (β=0.211, p<0.001), university students’ FA significantly and positively effected SPA (β=0.315, p<0.001), EI significantly predicted SPA in university students, and FA partially mediated the association between EI and SPA. The mediation effect of FA was 0.110, which accounted for 34.27% of the total effect. Bootstrap results furthermore tested the rigor of the mediating effect. @*Conclusion@#These findings broaden our understanding regarding the relationship between EI and SPA and the mediating role of FA, providing new sights for educators on how to reduce the risk of SPA when confronting the ongoing and possible future pandemics.

ABSTRACT

Objective To explore the expression of Per2 in hepatocellular carcinoma (HCC) and its effect on the survival of HCC patients, and to analyze the effects of Per2 on the proliferation and apoptosis of SMMC-7721 cells. Methods The protein expression of Per2 in HCC patients was analyzed by immunohistochemical (IHC) staining. SMMC-7721 cells were transfected with Per2 eukaryotic expression plasmid. The protein expression of Per2 was detected by Western blot. The cell proliferation and apoptosis was detected by MTS and flow cytometry. Results In 117 HCC tissues, the Per2 positive expression rate was 70. 94%, which was significantly lower than that of 87. 18% in the adjacent HCC tissue. The Per2 staining score was 2. 14±1. 76, which was significantly lower than that of 6. 39±3. 84 in the adjacent HCC tissue (P<0. 01). Per2 expression was related to tumor diameter, portal vein invasion and TNM staging(P<0.05) in 117 HCC patients. The overall survival (OS) and recurrence-free survival (RFS) of HCC pa-tients with low expression of Per2 were shorter than those with Per2 high expression(P<0.05).The cell proliferation was significantly inhibited and the apoptosis increased with the transfection of Per2 eukaryotic expression plasmid in SMMC-7721 cells(P<0.05).Conclusions The expression of Per2 in HCC is significantly reduced.Per2 inhibits the progression of HCC,possibly by inhibiting cell proliferation and promoting apoptosis.

ABSTRACT

Objective To investigate the epidemiological characteristics of shoulder injuries by training,and provide scientific evidence for effective intervention measures.Methods Through cluster random sampling method,a questionnaire survey was conducted on 1 080 trainers to analyze the occurrence of shoulder injury by training,the training courses lead to shoulder injuries and classification of shoulder injuries by training.The psychological status,shoulder symptoms,protective measures and other factors were compared between the injured group and the normal group to explore the influencing factors of injuries.Results Of 1 080 trainers,the incidence of shoulder injuries was 18.52%.The training course where injured were currently creeping forward (34.96%),push-ups (13.82%) and pull-ups (13.41%).The main types of injuries were soft tissue injuries(92.27%).The difference was statistically significant between two groups in the aspects of the mental state,shoulder protection measures and the presence of symptoms (P < 0.01).Conclusion The shoulder injuries of training personnel were mainly caused by soft tissue injuries.Long time training,short time for rest,lack of control or confidence were the major risk factors.Shoulder symptoms during training and inadequate application of protective measures may lead to the incidence of training injuries increasing.

ABSTRACT

Objective: To observe the effect of recombinant human bone morphogenetic protein 2/poly-lactide-co-glycolic acid (rhBMP-2/PLGA) with core decompression on repair of rabbit femoral head necrosis. Method: Bilateral femoral head necrosis models of rabbit were established by steroid injection. A total of 48 rabbits (96 femoral head necrosis) were randomly divided into 4 groups: Group A, control group with12 rabbits, 24 femoral head necrosis; Group B, treated with rhBMP-2/PLGA implantation after core depression, with 12 rabbits, 24 femoral head necrosis; Group C, treated with rhBMP-2 implantation after core depression, with 12 rabbits, 24 femoral head necrosis; Group D treated with core depression group without implantation, with 12 rabbits, 24 femoral head necrosis. All animals were sacrificed after 12 weeks. The ability of repairing bone defect was evaluated by X-ray radiograph. Bone mineral density analysis of the defect regions were used to evaluate the level of ossification. The morphologic change and bone formation was assessed by HE staining. The angiogenesis was evaluated by VEGF immunohistochemistry. Results: The osteogenetic ability and quality of femoral head necrosis in group B were better than those of other groups after 12 weeks by X-ray radiograph and morphologic investigation. And the angiogenesis in group B was better than other groups. Group C had similar osteogenetic quality of femoral head necrosis and angiogenesis with group D. Conclusion: The treatment of rhBMP-2/PLGA implantation after core depression can promote the repair of rabbit femoral head necrosis. It is a promising and efficient synthetic bone material to treat the femoral head necrosis.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the inhibitory effects of siRNA targeting BCSG1 gene expression in tumor transplants of human breast cancer cell line in nude mice.</p><p><b>METHODS</b>Four-pairs of small interfering RNA sequences of BCSG1 were chemically synthesized and inserted into the plasmid expression vectors, and were then transfected into human breast carcinoma cell line MCF7 by liposome method. Plasmid vector with unrelated sequence was used as the vector control. Cells transfected with 4 siRNA sequences, control vector and naive FCF7 cells were transplanted into the nude mice. The tumor inhibition was analysised. Immunohistochemical SP method and semi-quantitative RT-PCR were adopted to detect the BCSG1 mRNA and protein expression, respectively. Breast tissue samples of human infiltrating ductal carcinoma, ductal hyperplasia and fibroadenoma were also used as the controls.</p><p><b>RESULTS</b>The inhibition rates of tumor growth in four BCSG1-siRNA transfected groups were remarkably higher than those of the vector control group and naive MCF7 cells (P<0.01). Compared with that of the vector control and naïve MCF7 cell group, there was a significant decrease of BCSG-1 protein expression in the four experimental groups by immuno-histochemistry staining (P<0.01). In addition, BCSG1 mRNA expression in the four groups transfected with BCSG1-siRNA were significantly less than that of the control vector group, naive MCF7 cell control group and human breast IDC (P<0.01).</p><p><b>CONCLUSION</b>BCSG1-siRNA down-regulates the expression of BCSG1 and inhibits effectively growth of the transplaned human breast cancer cell line in nude mice.</p>

ABSTRACT

CTB protein possessed mucosal adjuvant immunoactivity. The CTB gene was amplified by PCR method from a strain V. cholerae. The nucleotide sequence of CTB gene was 375 bp and shared 96.0%~99.2% homology with other 6 CTB genes. The recombinant plasmid pTWIN1-CTB transformed E. coli strain BL21(DE3) expressed with 0.8 mmol/L IPTG. The molecular weight of expression products was identical with expectative weight by SDS-PAGE electrophoresis. The CTB fusion proteins mainly assembled inclusion bodies and the outputs of proteins were approximately 20% of the total bacterial proteins. The CTB proteins possessed mucosal immunoactivity by GM1-ELISA assay.

ABSTRACT

Objective Effectiveness of two kinds of thermochemotherapy infusion from intraarterial approach were studied in the grafted liver VX2 tumors of rabbit.Methods VX2 tumor model was established in 30 Newzland rabbit's livers.Percutaneous transfemoral hepatic arterial catheterization with fixation of the cathether tip inside the feeding vessel was carried out under DSA guidance.All 30 rabbits were divided into three groups(n=10 in each group),normal temperature 100 ml saline+Adriamycin(ADM)infusion(group 1),60℃100 ml saline+ADM continuous perfusion(group 2)and 60℃100 ml saline+ADM intermittent perfusion(group 3).After the perfusion,the lasting time periods of 43-45℃for tumor tissue of group 2 and 3 together with the concentrations of ADM within tumor's tissue were measured.Results Concentrations of ADM were shown as(12.013?2.237)?g/ml,(17.622?1.368)?g/ml,and(11.519?1.225)?g/ml for group 2, group 3 and group 1 respectively.60℃intermittent perfusion vs 60℃continuous perfusion showed P<0.05, 60℃continuous pefusion vs normal temperature perfusion also showed P>0.05. 43-45℃period lasting time (min)for 60℃continuous pefusion vs 60℃intermittent pefusion were(4.1?2.7)min and(11.3?3.3)min respectively,the latter was three times more than the former.There were no differences shown betwen the temperature,respiration and heart rate of group 2 and group 3.Conclusion Intermittent intraarterial perfusion thermochemotherapy is a more effective interventional management among all thermochemotherapies.

ABSTRACT

The Coat protein and Maturase gene of E.coli bacteriophage MS2 was amplified by PCR,then the gene was cloned into pET32a to construct the intermediate vector pET32aCP.The conservative sequence of FMDV internal ribosome entry site(IRES) was cloned into the downstream of pET32aCP bacteriophage gene to construct the prokaryotic expression vector pCPES.The recombinant plasmid pCPES transformed into E.coli strain BL21(DE3) was induced to express with 1mmol/L IPTG.The expression products were purified by sucrose density gradient centrifugation.The expression products observed by TEM were circular viruslike particles,and the diameter of these particles was about 26nm.The stability of viruslike particles was detected,and the viruslike particles was identified by RTPCR.The results showed that the viruslike particles contain the FMDV IRES RNA and have good stability.The viruslike particles have great prospect as the standard and quality control in the area of RNA virus detection.

ABSTRACT

Objective To explore the biological functions of retinoic acid-inducible gene-I(RIG-I) in vivo through phenotype analysis of RIG-I knockout mice. Methods The gene expression of RIG-Ⅰ in various tissues of mice was examined with Northern blotting and semi-quantitative RT-PCR.The phenotypes observed included body weight measurement,differential count of peripheral blood cells,metabolic parameters measurement and histopathologic examination. ResultsRIG-Ⅰ expressed in various tissues of mice with different levels.No gross developmental abnormalities and expected maturation arrest in granulocytic differentiation were observed in RIG-Ⅰ knockout mice.However,RIG-Ⅰ knockout mice exhibited an unexpected increase in the ratios of neutrophiles to lymphocytes in peripheral blood and increased susceptibility to bacteria infection. Conclusion RIG-Ⅰ may play an important role in immune regulation in mice.

Objective To evaluate the anti-tumour effect of 60℃ LipiodoI in the embolization of VX_2 hepatocarcinoma in rabbits.Methods VX_2 carcinoma cells were surgically implanted into the left liver lobe in 30 male New Zealand white rabbits,which were randomly divided into 3 groups by figure and table method with 10 rabbits in each group.Physiological saline,Lipiodol(37℃),and Lipiodol(60℃)were injected in each group via hepatic artery and liver cancer was embolized.The volume of tumour and serum level of aspartate aminotransferase(AST)were observed after one week,and the survival period of VX_2 rabbits was also observed.Results In the group of Lipiodol(60℃),the growth rate of tumour(0.92? 0.21)was significantly lower than that of control group(3.48?1.17)and Lipiodol(37℃)groups (1.69?0.26),respectively(F=34.95,P0.05),but was significantly higher than the control group(68.6?6.6)U/L(t=19.24,P
